Urothelial carcinoma (UC) originates from the urothelial lining renal pelvis, ureter, bladder, and urethra. Based on the report from Global Cancer Statistics 2020, bladder cancer is the 7th and 13th most frequent malignancy among men worldwide and those in Japan, respectively (1,2). Unresectable, metastatic, or advanced UC (aUC) is a progressive, poor prognostic disease with a median overall survival (OS) time of approximately 19–26 months following first-line (1L) platinum-based chemotherapy and subsequent administration of immune checkpoint inhibitors (ICIs) including anti-PD-1 and anti-PD-L1 antibodies (3-6). A substantial proportion of the population responds poorly to chemotherapy and ICIs, resulting in poor survival outcomes. Novel upfront drug combinations including ICIs, antibody-drug conjugates, and tyrosine kinase inhibitors (TKIs) are currently being tested in an attempt to overcome drug resistance and improve antitumor activity.
Controlling microstructure in fusion-based metal additive manufacturing (AM) remains a challenge due to numerous parameters directly impacting solidification conditions. Multiprincipal element alloys (MPEAs) offer a vast compositional design space for microstructural engineering due to their chemical complexity and exceptional properties. Here, we establish a novel alloy design paradigm in MPEAs for AM using the FeMnCoCr system. By exploiting the decreasing phase stability with increasing Mn content, we achieve notable grain refinement and breakdown of columnar grain growth. We combine thermodynamic modeling, operando synchrotron X-ray diffraction, multiscale microstructural characterization, and mechanical testing to gain insight into the solidification physics and its ramifications on the resulting microstructure. This work paves way for tailoring grain sizes through targeted manipulation of phase stability, thereby advancing microstructure control in AM.

The quest for targeted therapies is critical in the battle against cancer. The RAS/ MAP kinase pathway is frequently implicated in neoplasia, with ERK playing a crucial role as the most distal kinase in the RAS signaling cascade. Our previous research demonstrated that the interaction between ERK and MYD88, an adaptor protein in innate immunity, is crucial for RAS-dependent transforma- tion and cancer cell survival. In this study, we examine the biological con- sequences of disrupting the ERK-MYD88 interaction through the ERK D-recruitment site (DRS), while preserving ERK ' s kinase activity. Our results indicate that EI-52, a small-molecule benzimidazole targeting ERK-MYD88 interaction induces an HRI-mediated integrated stress response (ISR), resulting in immunogenic apoptosis speci fi c to cancer cells. Additionally, EI-52 exhibits anti-tumor ef fi cacy in patient-derived tumors and induces an anti-tumor T cell response in mice in vivo. These fi ndings suggest that inhibiting the ERK-MYD88 interaction may be a promising therapeutic approach in cancer treatment.
